    Cultivar considerations and pod sealants for straight-combining canola in Saskatchewan

    Non-Peer ReviewedWhile the recommended and preferred harvest practice in western Canada is to swath canola (Brassica napus L.), there is appreciable interest in straight-combining this crop. In a recent study, five cultivars were harvested according to one of four harvest treatments and evaluated for seed yield, yield loss due to shattering, percent green seed and seed size. The cultivars included four B. napus hybrids and an open-pollinated canola quality B. juncea variety. Harvest strategies were swathed, straight-combined without a pod sealant, straight-combined with Pod Ceal DC® and straight-combined with Pod-Stik®. While average yields ranged from 894-3066 kg ha-1, cultivar rankings for yield were generally consistent across sites. Seed yields were equal when averaged across harvest treatments and sites, but swathed yields differed from straight-combined yields 50% of the time for individual sites. At two sites, straight-combining produced 142-370 kg ha-1 higher yields than swathing while, when harvest was delayed due to unfavourable weather, swathed yields were 276-413 kg ha-1 higher. A 217 kg ha-1 yield increase occurred with pod sealants at one site, but there were no differences amongst the two products and pod sealants did not affect yields of straight-combined canola at the remaining seven sites. Pod sealants did not have a measurable effect on shattering losses, even under high shattering conditions. In contrast, cultivar effects on seed loss were generally significant with losses from one of the napus cultivars being particular and consistently low, especially when overall shattering losses were high. On average, losses for all cultivars were 4% of the total yield when harvest was completed reasonably close to the optimal stage. Straight-combining resulted in a small but significant increase in percent green seed and seed size but pod sealants did not affect seed quality in any cases. In conclusion, straight-combining can be a viable alternative to swathing, but substantial yield losses may occur if harvest is delayed too long. Important varietal differences in shattering losses were detected and cultivar selection appears to be a factor of greater importance than pod sealants for growers planning to straight-combine canola

    Flow profiling of a surface acoustic wave nanopump

    The flow profile in a capillary gap and the pumping efficiency of an acoustic micropump employing Surface Acoustic Waves is investigated both experimentally and theoretically. Such ultrasonic surface waves on a piezoelectric substrate strongly couple to a thin liquid layer and generate an internal streaming within the fluid. Such acoustic streaming can be used for controlled agitation during, e.g., microarray hybridization. We use fluorescence correlation spectroscopy and fluorescence microscopy as complementary tools to investigate the resulting flow profile. The velocity was found to depend on the applied power somewhat weaker than linearly and to decrease fast with the distance from the ultrasound generator on the chip.Comment: 12 pages 20 figure

    Signal peptide peptidases and gamma-secretase: Cousins of the same protease family?

    Signal peptide peptidase (SPIP) is an unusual aspartyl protease, which mediates clearance of signal peptides by proteolysis within the endoplasmic reticulum (ER). Like presenilins, which provide the proteolytically active subunit of the,gamma-secretase complex, SPP contains a conserved GxGD motif in its C-terminal domain which is critical for its activity. While SPIP is known to be an aspartyl protease of the GxGD type, several presenilin homologues/SPP-like proteins (PSHs/ SPPL) of unknown function have been identified by database searches. In contrast to SPP and SPPL3, which are both restricted to the endoplasmic reticulum, SPPL2b is targeted through the secretory pathway to endosomes/lysosomes. As suggested by the differential subcellular localization of SPPL2b and SPPL3 distinct phenotypes were found upon antisense gripNA-mediated knockdown in zebrafish. spp and sppl3 knockdowns in zebrafish result in cell death within the central nervous system, whereas reduction of sppl2b expression causes erythrocyte accumulation in an enlarged caudal vein. Moreover, expression of D/A mutants of the putative C-terminal active sites of spp, sppl2, and spp13 produced phenocopies of the respective knockdown phenotypes. These data suggest that all investigated PSHs/SPPLs are members of the novel family of GxGD aspartyl proteases. More recently, it was shown that SPPL2b utilizes multiple intramembrane cleavages to liberate the TNF(x intracellular domain into the cytosol and to release the C-terminal counterpart into the lumen. These findings suggest common principles of intramembrane proteolysis by GxGD type aspartyl proteases.     Microstructure, mechanical properties and fracture mechanisms in a 7017 aluminium alloy tailored for powder bed fusion – laser beam

    This study addressed a 7017 Al-alloy tailored for powder bed fusion – laser beam (PBF-LB) process. The alloy was prepared by mixing 3 wt% Zr and 0.5 wt% TiC powder to standard pre-alloyed 7017 grade aluminium powder. This made printing of the alloys possible avoiding solidification cracking in the bulk and achieving high relative density (99.8 %). Such advanced alloys have significantly higher Young\u27s modulus (>80 GPa) than conventional Al-alloys (70–75 GPa), thus making them attractive for applications requiring high stiffness. The resulting microstructure in as-printed condition was rich in particles originating from admixed powders and primary precipitates/inclusions originating from the PBF-LB process. After performing a T6-like heat treatment designed for the PBF-LB process, the microstructure changed: Zr-nanoparticles and Fe- or Mg/Zn- containing precipitates formed thus providing 75 % increase in yield strength (from 254 MPa to 444 MPa) at the cost of decreasing ductility (∼20 % to ∼9 %). In-situ tensile testing combined with SXCT, and ex-situ tensile testing combined with fracture analysis confirmed that the fracture initiation in both conditions is highly dependent on defects originated during printing. However, cracks are deflected from decohesion around Zr-containing inclusions/precipitates embedded in the Al-matrix. This deflection is seen to improve the ductility of the material

    Learning to live in a smart home

    Smart homes promise to significantly enhance domestic comfort, convenience, security and leisure whilst simultaneously reducing energy use through optimized home energy management. Their ability to achieve these multiple aims rests fundamentally on how they are used by householders, yet very little is currently known about this topic. The few studies that have explored the use of smart homes have tended to focus on special-interest groups and be quite short-term. This paper reports on new in-depth qualitative data that explore the domestication of a range of smart home technologies in 10 households participating in a nine-month field trial. Four core themes emerge: (1) smart home technologies are both technically and socially disruptive; (2) smart homes require forms of adaptation and familiarization from householders that can limit their use; (3) learning to use smart home technologies is a demanding and time-consuming task for which there is currently very little support available; and (4) there is little evidence that smart home technologies will generate substantial energy savings and, indeed, there is a risk that they may generate forms of energy intensification. The paper concludes by discussing the implications of these findings for policy, design and further research

    Changes in rod and frame test scores recorded in schoolchildren during development--a longitudinal study.

    The Rod and Frame Test has been used to assess the degree to which subjects rely on the visual frame of reference to perceive vertical (visual field dependence-independence perceptual style). Early investigations found children exhibited a wide range of alignment errors, which reduced as they matured. These studies used a mechanical Rod and Frame system, and presented only mean values of grouped data. The current study also considered changes in individual performance. Changes in rod alignment accuracy in 419 school children were measured using a computer-based Rod and Frame test. Each child was tested at school Grade 2 and retested in Grades 4 and 6. The results confirmed that children displayed a wide range of alignment errors, which decreased with age but did not reach the expected adult values. Although most children showed a decrease in frame dependency over the 4 years of the study, almost 20% had increased alignment errors suggesting that they were becoming more frame-dependent. Plots of individual variation (SD) against mean error allowed the sample to be divided into 4 groups; the majority with small errors and SDs; a group with small SDs, but alignments clustering around the frame angle of 18°; a group showing large errors in the opposite direction to the frame tilt; and a small number with large SDs whose alignment appeared to be random. The errors in the last 3 groups could largely be explained by alignment of the rod to different aspects of the frame. At corresponding ages females exhibited larger alignment errors than males although this did not reach statistical significance. This study confirms that children rely more heavily on the visual frame of reference for processing spatial orientation cues. Most become less frame-dependent as they mature, but there are considerable individual differences
